
To the sediment you used to make the smears for special stain add approximately 10 ml. of 10% formalin. Add 2 - 3 ml. of ether or ethyl acetate depending on method used and the consistency of the stool sediment. Mix the suspension well and centrifuge at 500 g. for 10 minutes. After centrifugation there should be 3 distinct layers visible in the tube. The top layer containing most of the fat, debris and ether. The middle layer of very small suspended material and formalin and the bottom layer of concentrated stool.

There should really be only a few drops of concentrated stool sediment at the bottom of the tube at this point.

To loosen the plug of debris gently ring the top of the tube with an applicator stick and dump the contents into a suitable container for ether evaporation and disposal. Remove the sediment adhering to the sides of the tube with a cotton swab. Try to leave a little fluid with the sediment for mixing and wetmount preparation. If you have to saline or formalin may be added.
